For starters, you won't be able to set ownership and permissions on a fat32 file system, and software running under gnu/linux, as well as the distros themselves don't like that. Also, technically, zipslack ran on umsdos, a unix file system running on top of msdos. I suppose it would be possible to install and use a gnu/linux distro running on umsdos, which would of course be on a fat/fat32 file system, but if you wanted to do that, you'd have to be contented with using a 2.4.x kernel, unless someone has ported umsdos to the 2.6.x tree, and I'm just not aware of it.
