If it's com1 I really think it should be ttys0. You don't NEED a link, you can just use /dev/ttys0. I had a laptop once that linked /dev/mouse to /dev/ttys0 but it could have just as easily been /dev/ttys0. As to if you should reboot the machine...I'm not sure. Sorry, been about a year since I dealt with a serial mouse.
