Bill, a lot of thanks!

I was experimenting and managed to solve the problem and just now I found that it is exactly as you said!

Running the command with:
--play-file=applause.wav --auto-play, sends the RTP packets!

Although, running like this (which I need for my test):
--play-file=applause.wav --auto-play --duration=30, seems to have the same problem as before!

As can someone assume, it was solved by applying the auto-play last (so priority matters?):
--duration=30 --play-file=applause.wav --auto-play

Followup: the pjsip --help doc says --auto-play only works for incoming calls, but browsing the code it looks like it should work for all calls. Try including --auto-play option, if that doesn't work then you'll have to step through the code and see what changes are needed to make auto-play work for outgoing calls.
