Actually, it DOES support flock as long as fuse utilizes it, and the GLFS version of fuse do (the glusterfs devs patched it in). As long as you are using the GlusterFS version of fuse, locking should work fine.

Székelyi, are using the GlusterFS provided fuse?

I'm sorry that I cannot answer your question, but do you have a choice in
what mail server to use? Those that use the Maildir format (Like for
example Postfix together with Courier-IMAP) does not need locking.
