I'd suggest throwing it into %{_optflags}.

FWIW, all KDE software is already built with -D_FILE_OFFSET_BITS=64, 
FindKDE4Internal.cmake forces it (unless off_t is already 64-bit by 
default). (Of course, this doesn't apply to external dependencies, such as 
virtuoso-opensource, which unfortunately shows up in the list of broken 
packages.)
