Does Changing Cache_dir location require a "Rebuild" of the cache?

That is pretty much the question I have:
If the Proxy has been running for a while and caching data to say:

*cache_dir ufs /var/spool/squid3 20480 16 256*

Does Changing Cache_dir location(s) to something like this

*cache_dir ufs /var/spool/squid3_cache-1 5120 16 256
cache_dir ufs /var/spool/squid3_cache-2 5120 16 256
cache_dir ufs /var/spool/squid3_cache-3 5120 16 256
cache_dir ufs /var/spool/squid3_cache-4 5120 16 256*

. . . require a "Rebuild" of the cache or cache table, or will the system
realize that it has changed and start all over again automatically?

IF there was data already in the 
*cache_dir ufs /var/spool/squid3 20480 16 256*
directory when you made the switch, IF you were to change the cache_dir back
to this location, would squid3 recognize that there is data there and carry
on from there OR would it wipe it out and start fresh again regardless of
what is there?

I realize it is probably a weird question, but I am just trying to
understand how it works and what it is thinking when I make changes.
