My somewhat old information is that for a MySQL based system one should restrict access to the application while the backup is made. Faster machines have reduced the time taken, but if the volume of data was to increase dramatically so would the time taken to back up? Some of my own systems can take several minutes to create the backup file which is still in the gigabytes after backing up but I don't have to stop web access as the backup will be a snapshot of the data at the time the process started.
