Anything can be a backing file for the file/mass storage gadget. However, if you configure file/mass storage gadget to pretend it is CD-ROM, you probably also intend for it to be used as CD-ROM, ie. use a file system designed for various types of CD-ROMs, eg. ISO9660 or UDF.
