systemctl start bluetooth
systemctl enable bluetooth # if you want it to run when the system is booted

Having done this, under Arch Linux I get the following:
[jason@jpc ~]$ bluetoothctl
[NEW] Controller 00:1F:E1:E9:4E:67 jpc.jasonjgw.net [default]
[bluetooth]# help
Available commands:
[...]

You are correct, however, that without the daemon running, no keystrokes are
accepted by bluetoothctl (whether or not Speakup is active).
