Brltty comes up much later in the boot process, and can happily deal with pretty much any port you throw at it. I've even used it once over a USB to serial converter, but it was flaky in that configuration. Since Speakup talks to the hardware directly, it's really best to get Speakup working on the built-in serial port, as you are much more likely to succeed.
