Yes as long as the two internal networks are on two separate LAN cards or network segments (which they seem to be). There is a bug in the current v2.2.5 which did not allow proper proxying of media between two internal networks. This has now been fixed however you will need to wait for the next version to be released in the next week or so. In the new version, the behaviour you need will be default and the Internal LANs are auto detected and all that is required is to enable the proxy in the INI. Only the media requiring proxying LAN1 to LAN2, LAN1 to Internet, LAN2 to Internet are proxied, all other internal calls on each LAN are not.
