you can look here for a jack guide:

Didn't help, JACK just crashes after starting

Post the output of jack here and we'll go from there.

If you run alsa-info.sh from the command line and post the output it
will help us determine what hardware you are using

While we are troubleshoot, would be nice if someone can send me a wav
file of the D-flat pitch. That will at least get me started on my
arrangement. Cheers

thanks

While I look at the jack issue you can generate a tone through alsa/pulse

For Db you could try :

speaker-test -f 277.183 -t sine

This should work with pulseaudio running, otherwise you will have to specify the card with the -D parameter

In your case this should work

pulseaudio -k; speaker-test -f 277.183 -t sine -Dhw:0

for me it is

pulseaudio -k; speaker-test -f 277.183 -t sine -Dplughw:0,0

For other octaves see http://en.wikipedia.org/wiki/D%E2%99%AD_%28musical_note%29
