do you have brltty configured to use /dev/ttyS3?
The default is /dev/ttyS0. In the ./configure, you would do
./configure --with-braille-device=/dev/ttyS3

Also, depending on how these serial ports are set up, do you have any
possibility of something on another serial port that is competing for the use of
/dev/ttys3? Kind of unusual that you are using the fourth serial port, but that
should still work. By the way, there's no problem using speakup and brltty
together, with different serial ports and different synth/display equipment.
