Perhaps _installing_ wine (which is properly done as root) should create an empty, read-only _file_ /root/.wine, so that any subsequent attempt to _run_ wine as root would fail because the prefix directory doesn't exist and can't be created. Granted this can't prevent running as root by specifying a different prefix.
