This depends almost entirely on what you mean. Can you describe what
multi-tenant means to you?

I think you're forced to use two squid instances. Then you could look into cache_peer directive to reduce content duplication between the two caches. My 2 cents, as unfortunately I'm not familiar with squid peers setups...
