If you are using linux, then the answer is yes to both questions.
You need to setup rules in your firewall to allow this type of traffic,
then you need DNS resolution for the clients using either internal or
external DNS servers and you need to source NAT the internal addresses
like 192.168.x.x. to your external IP address. But this is way off-topic
for the squid mailing lsit and there are a lot of instructions out there
how to do that.
You should probably disallow direct port 80 connections through your
firewall or the beenfit of your proxy will be very low as people will
start going direct ....
