Configure workstations to not let people change the proxy, in Microsoft Windows it is possible via editing Group Policy(Using Group Policy Editor). And another one is, to use your firewall to forbid or redirect the requests that did not pass trough your proxy. As you mentioned that users don't need to set a proxy server in the internet explorer, so it seems you have squid proxy operating in transparent mode. So the machine that the Squid running on most likely is the internet gateway and you can do your purpose there. I mean if there is no firewall running on that machine use the lovely powerfull IPTABLES ( better to call Netfilter ).
