You can run all of this on one box if you want but you'll have to consider that, in that case, you won't be able to stop the mail server to fix a problem unless you're willing to stop the database, web and file server at the same time. You'll also have to partition your drive(s) so that one service going crazy won't impact the other ones. Also, you'll hit the limits of your cpu/ram/network card/drive faster if you run multiple services instead of having them on dedicated boxes.
