su root problem

I can no longer from user mode make su root in terminal.  su root gives 
password prompt, but the correct password is not acceptet. If I log 
completely out  and log in as root it goes ok.
I have been experementing a lot with this box with among all setting 
different chmod permitions on lots of directories,  have a suspicion the 
problem is about that.

Wich file does the system read for password when I su root?
