You might be able to convince a filesystem to deal in larger block sizes, but the underlying block IO layer has a physical limit of PAGE_SIZE due to a somewhat arbitrary design restriction in the implementation of bios. Unless I've missed a patch and it's already possible, you could modify the kernel to do what you want but it won't do it "out of the box".
