Check with "fuser /dev/snd/pcmC0D0c" or similar. With Pd however a lot of people have problems using the RME cards directly. You may be luckier if you use Pd over Jack. Otherwise try to not use Pd's default device hw:0 when starting Pd but maybe use plughw:0 first or define a device in your ~/.asoundrc, that matches your favourite setup.
