Unless you set GDK_ENABLE_XFT to 0 explicitely, you are using the PangoXft backend then. Font configuration is done using fontconfig. Please check the fontconfig user documentation at http://pdx.freedesktop.org/~fontconfig/fontconfig-user.html You also mentioned that you are using GTK+-DirectFB. How fonts are configured depends on what version of PangoFT2 you are using for this. If you are using version 1.2, the font configuration is based on fontconfig as well.
