How are you deciding that G_THREADS_ENABLED is not defined? 
Does a test case like:

#include <glib.h>

#ifndef G_THREADS_ENABLED
#error "Threads were not enabled
#endif

int main ()
{
  return 0;
}
===

not compile succesfully?
