'man mke2fs' has all the details.  I usually do:

mke2fs -b 4096 -j -m 1 -O dir_index $DEVICE

It's not. It'll probably work, but perfect it isn't.

You should repartition first, if only to get the partitions right. Take a look at fdisk.
