The server seems not to be parsing the squiggle as a "home" indicator
and so thinks it's just a filename. Does the user exist on the
webserver?

Is this file really in
/path/to/server-root/etc/apache22/extra/httpd-userdir.conf or should it
be /etc/apache22/extra/httpd-userdir.conf?
Test that httpd-userdir.conf is really being loaded (put in a deliberate
syntax error and check it gets detected by configtest.
If all else fails, try the explicit form of UserDir:
UserDir /usr/users/*/public_html
which will map http://server/~chandler to
/usr/users/chandler/public_html.
