After a huge amount of to-ing and fro-ing, the details of which are too boring to relate, it turned out to be something devastatingly simple (isn't it always?). The scanner had somehow become assigned to the wrong IP address in my router's static DHCP table. What threw me was that it still worked as a printer *despite having the wrong IP number*. I can only assume that CUPS was finding it with some kind of broadcast or UPNP search, but that SANE needs more precise information. Wouldn't it be great if CUPS and SANE worked better together, or even merged?
